Greater Quint of Armor x3
Greater Mark of Armor x9
Greater Seal of Health x9
Greater Glyph of Mana Regeneration x3

Armor quints and reds are the best-in-slot runes for tanky supports because they give enough resistance to handle ADC poke. HP Quints and Magic Resist blues are standard for resistances, and I run 3 mana regen blues to aid Alistar's mana issues.
These are the standard masteries I run for all tanky supports, however it is a little weak for laning.
Against Mostly AD Teams
This setup is best when the enemy is heavy on AD, but you can still switch some things around in the defensive tree.
Against Mostly AP Teams
When the enemy has a lot of AOE magic damage, these masteries are optimal. If they have a lot of CC, look to take Oppression instead of 1 point in Perseverance.
Flash and Exhaust
Exhaust is usually taken as it scales extremely well in the late game. During the laning phase, Exhaust will allow you to disable/lower the enemies damage even more! Always remember to Exhaust early so that you can prevent the enemy from doing as much daamge as possible.
Flash and Ignite
You may want to opt for Flash and ignite if you are going to play extremely aggressive. This is an aggressive strategy, when paired with a duo lane partner can be effective for snowballing the lane, and pushing quickly and getting a fast tower. Combo's like Graves Alistar, or Trist Alistar are very powerful combinations that can burst an enemy in lane quickly. You should be able to 100-0 the enemy support or AD carry with a Flash Q-W and a full burst combo from your AD Carry.
