Greater Quint of Armor x3
Greater Mark of Attack Damage x9
Greater Seal of Health x9
Greater Glyph of Magic Resist x9
Attack damage marks are the most beneficial with Thresh’s kit as they allow you to do the most damage and they also increase the damage you do to minions, making it easier to push the wave to your advantage. Health and magic resist seals / glyphs are standard. Supports used to run Health Quints, but after the rune changes having Armor Quints and Health Seals are much more efficient.
These are the standard masteries I rune for Thresh, however it is a little weak for laning.
Against Mostly AD Teams
This setup is best when the enemy is heavy on AD, but you can still switch some things around in the defensive tree.
Against Mostly AP Teams
When the enemy has a lot of AOE magic damage, these masteries are optimal. If they have a lot of CC, look to take Oppression instead of 1 point in Perseverance.
Take ignite when the enemy team has a high dps support. Otherwise take exhaust, especially if they have a hypercarry/assassin.
